Inventi Rapid/Impact: Med Chem is the peer reviewed journal of Pharmaceutical Sciences. The journal considers the papers which relate chemistry, pharmacology and biology in drug designing and drug discovery and focuses on small organic molecules, computational chemistry, chemical biology and structural biology. SYNTHESIS AND STRUCTURAL CHARACTERIZATION OF NOVEL PYRIMIDINE DERIVATIVES VIA. BIGINELLI REACTION FOR THEIR ANTIMICROBIAL SPECTRUM

In 1893, the synthesis of functionalized 3,4-dihydropyrimidin-2(1H)-ones (DHPMS) via three-component condensation reaction of an aromatic aldehyde, urea and ethyl acetoacetate was reported for the first time by P. Biginelli. In the past decades, such Biginelli-type dihydropyrimidones have received a considerable amount of attention due to the interesting pharmacological properties associated with this heterocyclic scaffold. In this review oxazolo [3,2-a] and pyrimido [2,1-b][1,3]oxazine have been prepared from 3,4-dihydropyrimidin-2(1H)-ones.Then I was focused my work to make new derivatives of oxazolopyrimidine, and pyrimidooxazine, having sulfonyl linkage and to check their Antibacterial and Antifungal activity . DESIGN, SYNTHESIS AND BIOLOGICAL EVALUATION OF OXADIAZOLE DERIVATIVE AS ANTICONVULSANT AGENT

Keeping in view the importance of Oxadiazole and Azetidinone compound, it is planned to synthesize a hybrid molecule comprising Oxadiazole and Azetidinone and were evaluated for Anticonvulsant activity. The Structures of all the synthesized compounds were elucidated on the basis of physical characterization (Melting point, TLC) and different Spectroscopy techniques (IR, 1H NMR and Mass spectroscopy). In the present study, anticonvulsant activities of Oxadiazole derivatives were evaluated by maximal electroshock method (MES) at 30, 100 and 200 mg/kg dose levels. Neurotoxicity of the compounds was also assessed at the same dose levels. Synthesized Compounds (SK0101- SK0109) gives anticonvulsant activity at different dose when compare with standard drug Phenytoin among that SK0101, SK0103, SK0105 gives better activity then others. NEW GENERATION OF OXADIAZOLE DERIVATIVES AS POTENT ANTICONVULSANT

Epilepsy is one of such disease where selectively acting drugs are still lacking. Current drug therapy for epilepsy suffers from a number of disadvantages including the fact that the convulsions of approximately 25% epilepsies are inadequately controlled by medication. Oxadiazole have often been described as bio-isosters for amide & esters. Due to increase hydrolytic & metabolic stability of Oxadiazole ring improved pharmacokinetic & in-vivo performance is often observed, which makes these heterocyclic as important structural motif for the Pharmaceutical industry. Based on review of literature Oxadiazole is important compound for anticonvulsant activity, so this compound may be modified for better activities. “2-Benzylidene-2,3-dihydro-1H-inden-1-one” derivatives, is a bicyclic carbonyl compound possessing interesting biological activities ie., mast cell stabilising, anticoagulant, acetyl choline esterase inhibition, anti-inflammatory, anti-cancer, anti-malarial and antibacterial activities. Benzylidene substituted 9 Indanone derivatives have been synthesized. The structures of the newely synthesised compounds were established by spectroscopic interpretation (1H NMR, 13C NMR, Mass and IR spectra) studies. THE PYRAZOLONE: POTENT AND VERSATILE SMALL MOLECULE SCAFFOLD FOR KEY BIOLOGICAL TARGETS

The pyrazolone ring is an important structural moiety found in numerous pharmaceutically active compounds. This is mainly due to the ease preparation and the important versatile biological activity. When pyrazolones were discovered, they were mostly useful as a anti-inflammatory and analgesic but in recent times, they are known to exhibit antioxidant, anticancer, antibacterial and several other pharmacological actions like antifungal, protein-kinase inhibitor, anti inflammatory, analgesic, antipyretic, anticancer, anticonvulsant, ant diabetic, plant growth regulator ,and herbicidal. These derivatives were withdrawn from the market because of their adverse effects such as agranulocytosis, skin rashes and blood dyscrasis. So, structural modification of pyrazolones is necessary to minimize the side effect and to improve its therapeutic application. But, recently it again gain place in market due to their versatile actions especially in cardiovascular diseases.
