SIMULTANEOUS ESTIMATION OF NIMESULIDE, CHLORZOXAZONE AND PARACETAMOL BY HPLC AND UV SPECTROPHOTOMETRIC METHOD

ABSTRACT
Two novel methods are developed for simultaneous estimation of Paracetamol, Chlorzoxazone and Nimesulide in pure and dosage form. In method-I, A rapid, sensitive and economical high performance liquid chromatographic method for determination of Nimesulide, Chlorzoxazone and Paracetamol has been developed. The chromatographic system contained reverse phase C18 Column (250×4.6mm, 5µm) with a mixture of Acetonitrile: Water in ratio of 60:40 as a mobile phase at 1.0 ml min-1 flow. The pH of water in the mobile phase was adjusted to 3 by means of o-phosphoric acid (OPA). The method was validated as per ICH guidelines. The retention time of Paracetamol was 2.5 min, Chlorzoxazone was 4.1 min and Nimesulide was 7.1 min. and the calibration curve was linear in the range of 10-50, 32.5-162.5, 37.5-187.5 µg/mlfor Nimesulide, Paracetamol and Chlorzoxazone respectively. The percentage RSD for precision and accuracy of method was found to be less than 2 %. The lower limit of detection and limit of quantitation was within the limits. In method-II, simple, accurate, economical and reproducible UV spectrophotometric method for simultaneous estimation of Nimesulide, Chlorzoxazone and Paracetamol in combined dosage form has been developed. The 0.1N NaOH solution was used as common solvent. The present method employs multi-wavelength spectroscopic estimationat 257nm, 244nm and 223nm. Results of analysis were validated statistically and by recovery studies.
