UV-VIS SPECTROPHOTOMETRIC METHOD DEVELOPMENT AND VALIDATION FOR SIMULTANEOUS ESTIMATION OF AMLODIPINE BESYLATE, OLMESARTAN MEDOXOMIL AND HYDROCHLOROTHIAZIDE

ABSTRACT
New feasible, sensitive and economic methods of spectrophotometric estimation have been developed for the simultaneous quantitative determination of amlodipine besylate (AML), olmesartan medoxomil (OLM) and hydrochlorothiazide (HCZ). These drugs are practically water-insoluble. Hence, AML, OLM and HCZ, stock solution was prepared in azeotropic mixture (methanol: water 40:60 v/v). The pharmaceutical marketed formulation of above combination was estimated with the help of derivative ratio spectra zero crossing spectrophotometry method (I), double divisor ratio spectra derivative method (II), and multi-wavelength or multicomponent spectroscopy method (III). AML, OLM and HCZ showed maximum absorbance at 239 nm, 256 nm and 271 nm respectively. Beer’s law was obeyed in the concentration range 4-20 µg/mL, 4-20 µg/mL, 3-11 µg/mL respectively. All the three methods was validated in terms of linearity (R2=0.999, 0.999, 0.999 for method I, 0.998, 0.997, 0.989 for method II, and 0.998, 0.999, 0.998 for method III, precision in terms of % Relative Standard deviation (R.S.D.) for intraday and interday was within 0.094 % –1.471 % and 0.138%–1.514 % respectively), accuracy (within the limit) and specificity. The methods were found to be robust to variation in solvent system (40:60 ± 2 v/v) and absorbance due to variation in wavelength (?max ± 2). These methods are simple, precise, accurate, sensitive and reproducible and can be used for the routine quality control testing for the marketed pharmaceutical formulations.
